{"id":165468,"date":"2024-10-17T13:13:56","date_gmt":"2024-10-17T19:13:56","guid":{"rendered":"https:\/\/tecscience.tec.mx\/en\/?p=165468"},"modified":"2024-12-05T11:15:05","modified_gmt":"2024-12-05T17:15:05","slug":"tumor-on-a-chip","status":"publish","type":"post","link":"https:\/\/tecscience.tec.mx\/en\/biotechnology\/tumor-on-a-chip\/","title":{"rendered":"Tumors on a Chip Can Revolutionize Personalized Cancer Treatment"},"content":{"rendered":"\n<p>Reproducing the behavior of <strong>a tumor on a 3D-printed chip<\/strong>, measuring less than a quarter dollar coin in size, by cultivating its cells at a microscale and assessing the effects of various drugs, may pave the way for<strong> <a href=\"\/en\/human-social\/floyd-chilton-personalized-diets\/\" target=\"_blank\" rel=\"noreferrer noopener\">personalized medicine<\/a><\/strong> for cancer patients.<\/p>\n\n\n\n<p>For individuals diagnosed with cancer, <strong><a href=\"\/en\/science-communication\/cancer-en-un-chip-innovacion-biomedica-anticancer\/\" target=\"_blank\" rel=\"noreferrer noopener\">tumor-on-a-chip (ToC)<\/a><\/strong> technology could not only reduce costs in the search for a cure but also provide <strong>tailored treatments<\/strong> that meet their specific needs. This approach allows for drugs that more precisely target cancerous tissue while <strong>minimizing adverse effects<\/strong>, according to <a href=\"https:\/\/www.researchgate.net\/profile\/Salvador-Gallegos-Martinez\" target=\"_blank\" rel=\"noreferrer noopener\">Salvador Gallegos<\/a>. During his doctoral studies in <a href=\"https:\/\/maestriasydiplomados.tec.mx\/posgrados\/doctorado-en-biotecnologia?locale=en\" target=\"_blank\" rel=\"noreferrer noopener\">Biotechnology<\/a> at Tec, and now as a researcher at Harvard, he has worked extensively with these devices.<\/p>\n\n\n\n<p>Tumor-on-a-chip systems serve as <strong><a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/38866003\/\" target=\"_blank\" rel=\"noreferrer noopener\">valuable platforms for studying the physiology of cancerous tissues<\/a> <\/strong>and evaluating the efficacy and toxicity of treatments. \u201cIn the literature, this technology is also referred to as microphysiological systems. (Courtesy: Salvador Gallegos)<\/figcaption><\/figure>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Tumor on a Chip: A Path to Personalized Medicine<\/strong><\/h2>\n\n\n\n<p>At the end of 2020, the U.S. government approved the <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC10617761\/\" target=\"_blank\" rel=\"noreferrer noopener\">FDA Modernization Act 2.0<\/a>, which includes the use of <strong>organ-on-a-chip<\/strong> technology as an alternative to animal testing in drug evaluation.<\/p>\n\n\n\n<p>Gallegos notes that alongside testing on mice, rabbits, and other animals, there are also <strong>2D culture tests<\/strong>, where cells are grown on single-layer plastic plates. However, neither of these methods accurately mimics the behavior occurring in the human body.<\/p>\n\n\n\n<p>\u201cWhen trying to extrapolate results from these platforms, discrepancies arise; they don\u2019t align. This is where the inspiration for a tumor-on-a-chip comes in: to create a <strong>more robust and predictable laboratory model for drug testing<\/strong>. This way, when we expose it to a patient, we <strong>can be more confident that it will be effective<\/strong>\u2014that\u2019s personalized medicine,\u201d Gallegos explains.<\/p>\n\n\n\n<p>To develop personalized treatments, <strong>it\u2019s necessary to obtain cells from patients through a biopsy<\/strong>. These cells are then cultured in a microfluidic system, suspended in a hydrogel \u2014a polymer with elasticity and porosity akin to natural tissues, allowing for oxygenation and nutrient uptake\u2014 before testing various drugs.<\/p>\n\n\n\n<p>The <a href=\"\/en\/biotechnology\/chaotic-printing\/\" target=\"_blank\" rel=\"noreferrer noopener\">organs<\/a> or <strong>tumors on chips are typically smaller than 30 millimeters<\/strong>, and their structural barriers are<strong> usually made from two types of materials<\/strong>: <strong>polymeric resins created with 3D printers<\/strong> and a <strong>polymer called PDMS (polydimethylsiloxane)<\/strong>. Both materials are transparent, enabling researchers to observe cellular activity, and are permeable to gases like oxygen and CO2, essential for cell growth.<\/p>\n\n\n\n<p>\u201cWhile at Tec, I worked on <a href=\"\/en\/health\/artificial-intelligence-and-breast-cancer\/\" target=\"_blank\" rel=\"noreferrer noopener\">breast cancer<\/a>, directing fluid flows in a system that functions as a <a href=\"https:\/\/www.scopus.com\/record\/display.uri?eid=2-s2.0-85128801191&amp;origin=resultslist&amp;sort=plf-f&amp;src=s&amp;sid=28044790e539f71f781b86eaa9ffdbf6&amp;sot=aut&amp;sdt=a&amp;sl=18&amp;s=AU-ID%2857637390700%29&amp;relpos=5&amp;citeCnt=7&amp;searchTerm=\" target=\"_blank\" rel=\"noreferrer noopener\"><strong>microscopic reacto<\/strong><\/a><a href=\"https:\/\/www.scopus.com\/record\/display.uri?eid=2-s2.0-85128801191&amp;origin=resultslist&amp;sort=plf-f&amp;src=s&amp;sid=28044790e539f71f781b86eaa9ffdbf6&amp;sot=aut&amp;sdt=a&amp;sl=18&amp;s=AU-ID%2857637390700%29&amp;relpos=5&amp;citeCnt=7&amp;searchTerm=\"><strong>r<\/strong><\/a>. It has a cavity \u2014like a small pool\u2014 where the cells are located, with two channels on the sides: one for feeding, through which nutrients, oxygen, sugars, and growth factors flow, and another for purging waste. <strong>This setup mimics the blood flow and how a tissue or tumor grows outside the body<\/strong>,\u201d Gallegos explains.<\/p>\n\n\n\n<p>As part of the <a href=\"https:\/\/www.alvareztrujillolab.com\/\" target=\"_blank\" rel=\"noreferrer noopener\">Alvarez-Trujillo Lab<\/a>, he tested drugs like doxorubicin and docetaxel, <strong>evaluating drug transport, cell viability, toxicity, glucose consumption, and cell death<\/strong>.<\/p>\n\n\n\n<p>Currently, the research group is leveraging tumor-on-a-chip technology to study other types of cancer, including <a href=\"https:\/\/www.scopus.com\/record\/display.uri?origin=recordpage&amp;zone=relatedDocuments&amp;eid=2-s2.0-85160794738&amp;relpos=0\" target=\"_blank\" rel=\"noreferrer noopener\">colorectal<\/a>, lung, and prostate cancers.<\/p>\n\n\n\n<p><strong>Testing drugs on an organ-on-a-chip has no ethical implications<\/strong>, Gallegos states, as evaluations are conducted on independent cells created in the laboratory rather than within a human body. (Courtesy: Salvador Gallegos)<\/figcaption><\/figure>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Benefits for Patients and Pharmaceutical Companies<\/strong><\/h2>\n\n\n\n<p>The development of tumor-on-a-chip technology could mean <strong>reduced treatment costs <\/strong>for patients, says Gallegos. \u201cThe primary investment is upfront, for designing the analysis and validation. Once that&#8217;s established, the rest is financially accessible. <strong>With the platform ready, you only need to culture the cells and run multiple tests simultaneously<\/strong>, much like what was done with <a href=\"\/en\/tag\/covid-19\/\" target=\"_blank\" rel=\"noreferrer noopener\">COVID<\/a> detection.\u201d<\/p>\n\n\n\n<p>By using cells from the patient and considering their physiological, chemical, and genetic characteristics, <strong>it becomes possible to personalize treatments with drugs that optimally target specific types of cancer<\/strong>.<\/p>\n\n\n\n<p>In turn, this approach <strong>could reduce side effects and the time required to test one or more drugs<\/strong> compared to traditional cancer treatment methods.<\/p>\n\n\n\n<p>\u201cI recently read an article about a pilot study where <strong>cancer patients donated their biopsies, and tumors-on-a-chip were created using their cells<\/strong> to be exposed to drugs for evaluation. These were patients in advanced stages of cancer, and it was found that <strong>nearly 47% of them saw an increase in life expectancy of five to ten years<\/strong>.\u201d<\/p>\n\n\n\n<p>The time required to conduct drug tests on cancer-on-a-chip platforms varies; however, the average duration<strong> to determine whether a drug is toxic\u2014meaning it does not kill healthy cells or promote tumor growth\u2014typically takes about two weeks<\/strong>.<\/p>\n\n\n\n<p>The researcher believes that <strong>for pharmaceutical companies, these platforms also represent millions in savings<\/strong> when conducting clinical trials and research to validate drug efficacy. In fact, some companies are beginning to produce these devices at scale, though they face the challenge of customization to meet the specific needs of patients.<\/p>\n\n\n\n<p>Currently, researchers working with organ- or tumor-on-a-chip technology encounter several challenges. One is <strong>gaining the trust of physicians to adopt these devices<\/strong> in defining oncology treatments. Another involves <strong>achieving interconnectivity among various chips<\/strong> to develop systems that can replicate human body functions.<\/p>\n\n\n\n<p>\u201cWe will observe <strong>how the organ chips behave when connected with one another<\/strong>, assessing whether they resemble a living system. This is one of the main challenges: <strong>creating a biological entity that can grow in the lab<\/strong> to model diseases and evaluate drugs,\u201d he comments.<\/p>\n\n\n\n<p><strong><em>Did you find this story interesting?
