However, little have they explained about the damage it can also do to our brain.\u00a0<\/p>\n\n\n\n<p>According to various studies, high cholesterol levels can cause memory loss, brain inflammation and other cognitive problems associated with\u00a0<strong>neurodegenerative diseases, such as\u00a0<a href=\"\/en?s=alzheimer%27s\" target=\"_blank\" rel=\"noreferrer noopener\">Alzheimer&#8217;s<\/a>.\u00a0<\/strong><\/p>\n\n\n\n<p>The mechanisms that explain how this happens are becoming increasingly clear thanks to a line of research that has been building for decades.<\/p>\n\n\n\n<p>One of its key moments was when\u00a0<a href=\"https:\/\/ki.se\/en\/people\/ingemar-bjorkhem\" target=\"_blank\" rel=\"noreferrer noopener\">Ingemar Bj\u00f6rkhem<\/a>, a physician and researcher at the Karolinska Institutet in Stockholm, demonstrated that\u00a0<strong>circulating cholesterol\u00a0can indeed cross the blood-brain barrier<\/strong> \u2013the structure that regulates the passage of substances and molecules from the blood into the brain. To do so, however, it must first be broken down into a smaller version called 27-hydroxycholesterol (27-OH).<\/p>\n\n\n\n<p>Once defined, researchers such as\u00a0<a href=\"https:\/\/ki.se\/en\/people\/angel-cedazo-minguez\" target=\"_blank\" rel=\"noreferrer noopener\">\u00c1ngel Cedazo Minguez<\/a>, also at Karolinska, continued to piece together the puzzle in order to understand how the abundance of this compound relates to neurodegeneration.<\/p>\n\n\n\n<p>&#8220;Then I came in, trying to find the molecular mechanisms through which this excess harms the brain,&#8221; says\u00a0<a href=\"https:\/\/mx.linkedin.com\/in\/raul-loera-valencia-36858941\" target=\"_blank\" rel=\"noreferrer noopener\">Ra\u00fal Loera<\/a>, a research professor at the\u00a0<a href=\"\/en\/tag\/school-of-medicine-and-health-sciences\/\" target=\"_blank\" rel=\"noreferrer noopener\">School of Medicine and Health Sciences<\/a>\u00a0(EMCS), Chihuahua Campus, of the Tecnol\u00f3gico de Monterrey, who completed his postdoctoral research at Karolinska.<\/p>\n\n\n\n<p>Loera has worked with transgenic animal models, cell cultures, and human studies to discover what happens to brain cells when there are large amounts of this lipid.<\/p>\n\n\n\n<figure class=\"wp-block-embed is-type-video is-provider-youtube wp-block-embed-youtube wp-embed-aspect-16-9 wp-has-aspect-ratio\"><div class=\"wp-block-embed__wrapper\">\n<iframe title=\"Conoce c\u00f3mo una dieta antiinflamatoria puede evitar el deterioro cognitivo\" width=\"800\" height=\"450\" src=\"https:\/\/www.youtube.com\/embed\/GsiyzDN1Hcc?feature=oembed\" frameborder=\"0\" allow=\"accelerometer; autoplay; clipboard-write; encrypted-media; gyroscope; picture-in-picture; web-share\" referrerpolicy=\"strict-origin-when-cross-origin\" allowfullscreen><\/iframe>\n<\/div><figcaption class=\"wp-element-caption\">English captions for &#8220;Discover How an Anti-Inflammatory Diet Can Help Prevent Cognitive Decline&#8221; video are available.<\/figcaption><\/figure>\n\n\n\n<h2 class=\"wp-block-heading\">Excess Cholesterol in the Brain in Laboratory Studies<\/h2>\n\n\n\n<p>In the human body, when cholesterol levels rise due to genetic causes, metabolic diseases, diet, or age, an imbalance occurs that eventually affects the brain.<\/p>\n\n\n\n<p>As this lipid increases, so does its 27-OH derivative, creating a gradient that pushes these molecules upward into the head.<\/p>\n\n\n\n<p>&#8220;Once inside, it starts interfering with important functions such as the connection between neurons, which correlates with Alzheimer&#8217;s disease,&#8221; says Loera.\u00a0<\/p>\n\n\n\n<p>To decipher this, the researchers\u00a0<a href=\"https:\/\/www.nature.com\/articles\/s41598-021-83008-3\" target=\"_blank\" rel=\"noreferrer noopener\">worked with a transgenic animal model<\/a>\u00a0with permanently elevated levels of 27-OH. In this model, neurons in the hippocampus, a brain region essential for forming memories, showed a reduction in dendritic spines\u2014the small protrusions where synapses form\u2014affecting the way they communicate.\u00a0<\/p>\n\n\n\n<p>&#8220;That loss of connections is the first memory loss that patients with this disease report,&#8221; the expert explains.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Glutamate Toxicity and Brain Inflammation<\/h2>\n\n\n\n<p>Another mechanism they found in neuronal cultures is that excess 27-OH is toxic to oligodendrocytes, brain cells that have various functions, such as forming myelin\u2014a layer made of lipids and proteins that hydrates, insulates, and protects nerves and neurons.<\/p>\n\n\n\n<p>They also found that it <a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/30395175\/\" target=\"_blank\" rel=\"noreferrer noopener\">damages the shape and function of\u00a0hippocampal\u00a0neurons<\/a> and\u00a0<a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/34449045\/\" target=\"_blank\" rel=\"noreferrer noopener\">generates brain inflammation<\/a>, affecting their performance.<\/p>\n\n\n\n<p>In animal models, researches <a href=\"https:\/\/www.nature.com\/articles\/s41380-021-01257-0\" target=\"_blank\" rel=\"noreferrer noopener\">found<\/a>\u00a0that astrocytes\u2014the brain cells that support and protect neurons\u2014stop fulfilling their function of clearing up glutamate, a neurotransmitter crucial to learning and memory.<\/p>\n\n\n\n<p>&#8220;The excessive glutamate accumulation is another sign of stress in the brain,&#8221; says\u00a0<a href=\"https:\/\/ki.se\/personer\/luis-enrique-arroyogarcia\" target=\"_blank\" rel=\"noreferrer noopener\">Luis Enrique Arroyo<\/a>, a researcher at Karolinska.\u00a0<\/p>\n\n\n\n<p>To confirm the link between excess 27-OH and memory loss, the researchers\u00a0used a memory test\u00a0called the Morris water maze in animal models and observed that those with elevated amounts of this derivative had difficulty remembering the location of a platform that was submerged underwater, compared to those who did not have this excess and could remember it.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Does this Happen in Humans Too?<\/h2>\n\n\n\n<p>Beyond laboratory experiments, Loera and his colleagues have conducted studies with humans to see if what they&#8217;ve found applies to us.<\/p>\n\n\n\n<p>&#8220;We&#8217;ve studied it in blood, serum, cerebrospinal fluid, and tissue samples from deceased patients,&#8221; says Loera. They found that people with Alzheimer&#8217;s or cognitive impairment had elevated levels of 27-OH.<\/p>\n\n\n\n<p>In an epidemiological study\u00a0<a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/23332672\/\" target=\"_blank\" rel=\"noreferrer noopener\">called FINGER<\/a>, led by\u00a0<a href=\"https:\/\/ki.se\/en\/people\/miia-kivipelto\" target=\"_blank\" rel=\"noreferrer noopener\">Miia Kivipelto<\/a>\u00a0of Karolinska, they measured levels of this compound in older adults with Alzheimer&#8217;s or cognitive impairment.<\/p>\n\n\n\n<p>\u201cAccording to Miia\u2019s findings, when there is hypercholesterolemia (high cholesterol), people&nbsp;<strong>develop Alzheimer\u2019s earlier and it progresses more quickly<\/strong>,\u201d says Loera.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">What Does This Mean for the Prevention and Treatment of Neurodegenerative Diseases?<\/h2>\n\n\n\n<p>With all this in mind, one would think the solution would be as simple as lowering cholesterol levels in the body, either by modifying the diet or taking medication. Unfortunately, researchers have found that this is not an effective strategy.<\/p>\n\n\n\n<p>&#8220;When you lower cholesterol or clear 27-OH from the periphery,\u00a0<strong>it doesn&#8217;t leave the brain \u2013it stays elevated<\/strong>,&#8221; Loera says. For now, scientists are still searching for an explanation.\u00a0<\/p>\n\n\n\n<p>Meanwhile, another one of Loera&#8217;s goals is to find therapeutic targets that can stop the conversion of cholesterol to 27-OH, thus preventing it from reaching the brain. He also seeks to find molecules that can be controlled with drugs or therapies to reduce their levels in the brain.<\/p>\n\n\n\n<p>Through an international collaboration between the Karolinska Institute and Tec de Monterrey, progress is expected soon in this line of research.<\/p>\n\n\n\n<p>For now, the message for preventing the development of Alzheimer&#8217;s or other neurodegenerative diseases \u2013in part\u2013 remains the same: <strong>to avoid high blood cholesterol levels by exercising and maintaining a healthy diet.<\/strong><\/p>\n\n\n\n<p>However, Loera stresses that it&#8217;s not about completely avoiding fat in our diet. Although excess cholesterol can be harmful to the body and brain, its absence is also dangerous.<\/p>\n\n\n\n<p>Ultimately, cholesterol, along with other lipids and proteins, is essential for proper brain function. The important thing is to maintain a healthy balance, with neither too much of it nor too little.<\/p>\n\n\n\n<p>&#8220;To date, Alzheimer&#8217;s is virtually incurable, so we must promote lifestyle choices that prevent neurodegeneration,&#8221; says Loera.<\/p>\n\n\n\n<p><br><br><strong><em>Were you interested in this story?
